Full-timeDescription
Position Title:
Sr. Scientist/ Principal Scientist, Translational Development
Reports to:
Director, Translational Development
Company Background:
Century Therapeutics is an innovative biotechnology company that is building an industry-leading allogeneic iPSC-derived cell therapy platform that integrates cutting-edge gene editing, protein engineering, technical development, and manufacturing capabilities to generate allogeneic, iPSC-derived NK and T cell therapies for cancer and autoimmune and inflammatory diseases. Century’s foundational technology is built on induced pluripotent stem cells (iPSCs) that have unlimited self-renewing capacity.
This enables complex cellular engineering to produce controlled cell banks of modified cells that can be expanded and differentiated into desired immune effector cells, which can supply vast amounts of allogeneic, homogeneous therapeutic products. This platform differentiates Century from other approaches that utilize non-renewable donor-derived cell products for the treatment of cancer and autoimmune and inflammatory diseases with significant unmet medical needs.
Role:
The Century team is seeking a motivated cellular immunologist who will work within the Translational Development team and be responsible for developing, validating, and implementing, either in-house or at CROs, a suite of clinical biomarker assays for Century’s allogeneic NK and T cell studies. The individual in this position will have the exciting opportunity to interrogate clinical trial samples to understand the mechanism of action of our novel products. The role is expected to be 80-100% lab-based.
Responsibilities:
- Lead assay development, optimization, and establishment (including fit-for-purpose validation/ qualification). Quickly learn/adapt advances from the field and strive for scientific innovation
- Develop multicolor flow cytometry-based assays to monitor the pharmacodynamics of Century’s products in clinical specimens
- Develop assays for evaluating cellular immunogenicity against the product (i.e. NK and T cell functional assays including proliferation, degranulation, ELISspot, etc.)
- Develop and optimize assays to monitor humoral immunogenicity against the product
- Conduct and oversee timely clinical sample testing and delivery of good-quality results
- Hands-on testing of immunogenicity assays, flow phenotyping, and other biomarker assessments as required
- For outsourced assays, manage timelines and provide technical oversights to CRO as required
- Analyze emerging data, present results, and interpretation of translational data for Translational and Early Development team management
- Contribute to the translational/clinical biomarker strategies for the Century’s programs
- Collaborate with research, analytical development, QC, compliance, and computational biology teams as appropriate, to achieve the above objectives
Qualifications:
- MS plus 10+ years, PhD plus 4-8+ years of experience in immunology, cell biology/ oncology, or related field
Requirements:
- Strong verbal and written communication skills
- Strong background in immunology and understanding of concepts of immune rejection required
- Biomarker assay development & validation experience in an industry or related setting
- Critical thinking and excellent experimental design skills, strong problem-solving and assay troubleshooting ability
- Expertise with multi-parameter flow cytometry is a must. Experience with spectral flow platform(s) is a plus
- Experience with human T cell/ NK cell functional assays (ELISpot, MLRs, ADCC, etc.) Hands-on experience in isolation and culture of primary human cells from blood, marrow, and tumor
- Management experience in assay development work at CROs/external partners preferred
- Working knowledge of GxPs
Preferred Skills:
- Experience and understanding of oncology/I-O and/or autoimmune disease areas preferred
- Experience working with human serum and plasma for LBA (ELISA, MSD, Luminex, etc.) is a plus
- Experience with cellular and humoral immunogenicity assay development
- Experience presenting data to a diverse cross-functional audience
